500 Photodynamic therapy optimization after 5-Fluorouracil treatment in Squamous Cell Carcinoma

Non-melanoma skin cancer (NMSC) is the most common type of worldwide, being subdivided in two types: basal cell carcinoma (BCC) and squamous (SCC). SCC more aggressive than BCC, triggering severe health aesthetic complications. Among non-invasive therapies to treat SCC, Photodynamic therapy (PDT) 5-Fluorouracil (5-FU) are frequently employed clinic. PDT uses a photosensitizing agent which presence red light produces reactive oxygen species (ROS) causing death. 5-FU chemotherapeutic drug that inhibits thymidylate synthase, repressing cycle. Although satisfactory results generally obtained, some cells can survive, proliferate even disseminate, producing tumor relapses. In sense, we aim study role resistance development on evaluating efficacy modulation other therapies, focusing specifically PDT. To these processes, isolated resistant lines SCC13 CSZ exposing them 10 cycles 72-hour (10 μM). The viability assay MTT was used after each treatment confirm tested resistances. PpIX accumulation studied by flow cytometry as well its localization since it related with ROS production evaluated measuring fluorescence intensity addition, After, an evaluation measured 3D-spheroids. We observed line increased sensibility damage showing higher membrane lysosome production. opposite tendency shown. As conclusion, pretreatment favors 2D 3D cultures but lower BCC. further studies needed, optimizes models.
